Application to Database server connection protocol

My web application is hosted into a server and my database for that web application is hosted into another server.

Say, I made a DB intensive request to web server. I know a request to the web server establishes an HTTP connection.

But I would like to know, what protocol is used for making a web server to DB server connection for that particular or any HTTP request??
